Given its debut at the Geneva Salon during March 1966, Lamborghini's 400 GT 2+2 featured a number of important differences compared to the two-seater variants it replaced. Mechanically, the 2+2 dropped ZF's five-speed gearbox in favour of a smoother and considerably quieter Lamborghini-designed five-speed transmission (this also having reputedly been seen in a handful of late 400 GT's). There was also an in-house differential to replace what had been a Salisbury item beforehand. The switch to steel bodies was now complete for reasons of manufacturing ease and added durability, just the bootlid and bonnet remaining lightweight alloy items.
 
To increase rear headroom, Touring subtly lowered the rear floorpan, raised the roofline and positioned the body slightly higher on the chassis, raising overall height by just over 2½-inches compared to the outgoing two-seaters. The rear windscreen was significantly reduced in size to maximise boot space and with its increased practicality, the 400 GT 2+2 proved almost twice as popular as earlier models, total production reaching a very respectable 222 units, five of which were right-hand drive.
